Intruduction: | The protein encoded by human matrix Gla protein (MGP) gene is secreted and likely acts as an inhibitor of bone formation. The encoded protein is found in the organic matrix of bone and cartilage, thought to act as an inhibitor of bone formation. Defects in this gene are a cause of Keutel syndrome (KS). Two transcript variants encoding different isoforms have been found for this gene. Extracellular domain of human MGP cDNA (20 - 96aa) was constructed with codon optimization and with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal. This protein was expressed in E. coli as inclusion bodies, refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
