Intruduction: | The Drosophila embryonic protein snail is a zinc finger transcriptional repressor which down-regulates the expression of ectodermal genes within the mesoderm. The nuclear protein encoded by this human SNAL1 gene is structurally similar to the Drosophila snail protein, and is also thought to be critical for mesoderm formation in the developing embryo. Recent data also demonstrated that SNAL1 plays important roles in Epithelial to Mesenchymal transition (EMT) and maintenance of embryonic mesoderm. Full-length human SNAL1 cDNA (264aa) was constructed with codon optimization and with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal. This protein was expressed in E. coli as inclusion bodies, refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
