Intruduction: | Human NRF2 gene encodes a transcription factor which is a member of a small family of basic leucine zipper (bZIP) proteins. NRF2 binds to antioxidant response (ARE) elements in the promoter regions of target genes. It is important for the coordinated up-regulation of genes in response to oxidative stress. NRF2 may be involved in the transcriptional activation of genes of the beta-globin cluster by mediating enhancer activity of hypersensitive site 2 of the beta-globin locus control region. This proteins may also be involved in response to injury and inflammation which includes the production of free radicals. Full-length human NRF2 cDNA (604aa, Isoform-I, derived from BC011558) was constructed with codon optimization using gene synthesis technology and expressed with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal. It was expressed in E. coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
