Intruduction: | Human Sox5 gene encodes a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. The Sox5 may act as a transcriptional regulator after forming a protein complex with other proteins. It may play a role in chondrogenesis. Recent data indicated that SOX5 can activate the HMGB2 enhancer for subsequent gero-protective effects such as rejuvenation of aged knee joints through epigenetic remodeling (over-expression of human Sox5 gene) . Full-length human Sox5 cDNA (762aa, Isoform-I, derived from BC060773) was constructed with codon optimization using gene synthesis technology and expressed with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal and 11 arginine (11R) tag at its C-terminal. It was expressed in E. coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
