| Intruduction: | This gene encodes a nuclear-encoded mitochondrial bifunctional enzyme with methylenetetrahydrofolate dehydrogenase and methenyltetrahydrofolate cyclohydrolase (MTHFD2) activities. The enzyme functions as a homodimer and is unique in its absolute requirement for magnesium and inorganic phosphate. Formation of the enzyme-magnesium complex allows binding of NAD. Recent data indicated that this enzyme activity plays an important role in T cell function for AIDS progress and tumor development. Full-length human MTHFD2 (30 – 350aa) gene was constructed with 15 N-terminal T7 tag. This protein was expressed in E. coli as inclusion bodies, refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
