Intruduction: | Human succinyl-CoA:3-ketoacid coenzyme A transferase 1, mitochondrial (OXCT1) gene encodes a member of the 3-oxoacid CoA-transferase gene family. The encoded protein is a homodimeric mitochondrial matrix enzyme that plays a central role in extrahepatic ketone body catabolism by catalyzing the reversible transfer of coenzyme A from succinyl-CoA to acetoacetate. Mutations in this gene are associated with succinyl CoA:3-oxoacid CoA transferase deficiency. It has been demonstrate that anti-OXCT1 auto-antibody could be detected in some auto-immuno-disease patients blood samples. Full-length mature human OXCT1 cDNA (40 - 520aa, derived from BC009001) was constructed with codon optimization and expressed with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal. This protein is expressed in E. coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
