Intruduction: | Human mediator of RNA polymerase II transcription subunit 6 (MED6) is a component of the Mediator complex, a co-activator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional pre-initiation complex with RNA polymerase II and the general transcription factors. Auto-antibody for MED6 has been detected in renal-cell carcinoma patients. Full-length human MED6 ( 157aa, Isoform-II ) gene was constructed with 15 aa N-terminal T7 Tag and expressed in E.coli as inclusion bodies. The protein was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
