Tumor necrosis factor alpha, (TNFα) is a cell signaling protein, which involved in systemic inflammation and is one of the cytokines that make up the acute phase reaction. It is produced chiefly by activated macrophages, although it can be produced by many other cell types such as CD4+ lymphocytes, NK cells, neutrophils, mast cells, eosinophils, and neurons. The primary role of TNF is in the regulation of immune cells. TNF, being an endogenous pyrogen, is able to induce fever, apoptotic cell death, cachexia, inflammation and to inhibit tumorigenesis and viral replication and respond to sepsis via IL1 & IL6 producing cells. Anti-TNFa antibodies have been developed for clinical applications, such as treatment of rheumatoid arthritis, inflamaatory bowel diseases and psoriasis, et al. As research reagents, anti-human TNFa antibody could be used for its detection or functional blocker for animal testing of human TNFa activities.

Phage-display derived anti-human TNFa specific antibody (human FN3 backbone) gene was constructed with 29 aa N-terminal T7 / His / TEV cleavage site Tag and expressed in E.coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified.
