| Intruduction: | Human amelogenin, X isoforms (AMELX) gene encodes a member of the amelogenin Recombinant Human AMELX Protein family of extracellular matrix proteins. Amelogenins are involved in biomineralization during tooth enamel development. Mutations in this gene cause X-linked amelogenesis imperfecta. Full-length mature human AMELX cDNA (17–191aa, isoforms-1, derived from BC064379) was constructed with codon optimization and expressed with a human Alpha-Fetal Protein N-terminal domain (AFPn)-His-TEV cleavage site (219aa) fusion at its N-terminal. This protein is expressed in E.coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
