| Intruduction: | Glucosamine-6-phosphate isomerase 1 (GNPDA1) is an allosteric enzyme that catalyzes the reversible conversion of D-glucosamine-6-phosphate into D-fructose-6-phosphate and ammonium. Recent data indicated that GNPDA1 seems to trigger calcium oscillations in mammalian eggs. These oscillations serve as the essential trigger for egg activation and early development of the embryo. Full-length human GNPDA1 cDNA (2–289 aa) was constructed with codon optimization and expressed with a small T7-His-TEV cleavage site Tag (31aa) fusion at its N-terminal. This protein is expressed in E.coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified. |
